Coultreon Biopharma is advancing COL-5671, an oral, once-daily inhibitor of salt-inducible kinase 3 (SIK3). This mechanism simultaneously suppresses pro-inflammatory cytokines (IL-23 and TNFα) while upregulating the immunoregulatory molecule IL-10, potentially offering more durable disease control than existing single-pathway biologics. Originally developed by Galapagos, COL-5671 has demonstrated preclinical efficacy across a spectrum of immune-mediated conditions. The USD $125M Series A funding, backed by Sofinnova, Novo Holdings, and Forbion, is earmarked to advance the candidate into Phase 2 trials for psoriasis and ulcerative colitis.
With clinical proof-of-concept data expected by 2027, Coultreon enters a crowded but high-value immunology market currently dominated by massive capital inflows and M&A. While the 8-year regulatory data protection (RDP) baseline remains intact, the standard 2-year market protection has been halved to 1 year, with additional one-year extensions now contingent on meeting specific clinical or public health milestones.
Under the compromise texts published in March 2026, companies can "earn" back protection through three distinct pathways: addressing an unmet medical need, conducting comparative clinical trials in multiple Member States, or securing a new therapeutic indication with significant clinical benefit. Furthermore, a novel Transferable Exclusivity Voucher (TEV) for priority antimicrobials offers a 12-month RDP extension that can be applied to any centrally approved product in a firm's portfolio.

Eli Lilly has entered a strategic alliance with California-based startup Profluent to develop AI-designed recombinase editors.
